DNA replication (occurring in S-phase of the eukaryotic cell cycle) is the semiconservative copying of a DNA molecule, producing two daughter molecules each with one old and one new strand.

Definition: DNA replication is the biological process by which a DNA molecule produces two identical copies of itself, ensuring genetic information is faithfully passed from a parent cell to its daughter cells.

Phase of occurrence in Eukaryotes: DNA replication occurs during the S (Synthesis) phase of Interphase in the eukaryotic cell cycle — the chromosome number remains the same (2C to 4C DNA content) but the amount of DNA doubles, preparing the cell for division.

Mechanism of DNA replication:

  1. Unwinding: The enzyme helicase unwinds and separates the two strands of the parental DNA double helix at a point called the origin of replication, forming a Y-shaped replication fork. Single-strand binding proteins (SSBs) stabilise the separated strands, and topoisomerase relieves the supercoiling/torsional strain ahead of the fork.
  2. Priming: Since DNA polymerase cannot initiate synthesis on its own, the enzyme primase lays down a short RNA primer to provide a free 3'-OH end.
  3. Synthesis: DNA polymerase adds nucleotides complementary to the template strand, always in the 5' → 3' direction.
