Trisomy of chromosome 21 (an extra copy of chromosome 21) causes Down's syndrome, a chromosomal disorder arising from non-disjunction during gamete formation.

During meiosis in a parent, if homologous chromosome 21 fails to separate (non-disjunction) at anaphase, a gamete may end up with two copies of chromosome 21 instead of one. When such a gamete fuses with a normal gamete at fertilization, the resulting zygote has three copies of chromosome 21 (trisomy 21) instead of the usual two, giving a total of 47 chromosomes instead of 46. This condition is called Down's syndrome.
