No single culture medium supports optimum growth for every kind of plant tissue, so choosing and correctly preparing a medium is one of the biggest determinants of whether a tissue culture experiment succeeds. MS medium (Murashige and Skoog, 1962) is the medium most widely used across all kinds of plant tissue culture work; it supplies a defined set of macronutrients, micronutrients, minor nutrients, an iron stock, vitamins, growth hormones (auxins and cytokinins such as IAA and kinetin), myo-inositol and sucrose as a carbon source.
