Unlike a competitive inhibitor, a non-competitive inhibitor does not resemble the substrate and does not try to occupy the active site. Instead, it binds to a completely separate part of the enzyme molecule. This binding changes the overall three-dimensional shape of the enzyme, including the shape of the active site itself, so that even though the substrate may still be able to bind there, the enzyme can no longer carry out catalysis effectively. Because the inhibitor is not competing with the substrate for the same site, simply increasing the substrate concentration does not reverse a non-competitive inhibitor's effect, which distinguishes it clearly from competitive inhibition.
