Mechanism of action: Statins act as competitive inhibitors of the enzyme HMG-CoA reductase (3-hydroxy-3-methyl-glutaryl-CoA reductase). This enzyme normally catalyses a key, rate-limiting step in the body's endogenous pathway for synthesising cholesterol. Because the statin molecule structurally resembles the enzyme's natural substrate, it competes for and occupies the enzyme's active site, thereby blocking/reducing the enzyme's catalytic activity.
