Indirect ELISA detects whether a specific antibody is present in a patient's sample. First, a known antigen is fixed (coated) onto the surface of a test well. The patient's serum sample is then added; if the specific antibody being tested for is present, it binds to this fixed antigen. Anything unbound is washed away, and a second, "detecting" antibody — linked to an enzyme, and specifically recognising the patient's (first) antibody — is added and allowed to bind.
