Matthew Meselson and Franklin Stahl experimentally confirmed in 1958 that DNA replicates semi-conservatively, i.e. each daughter DNA molecule retains one parental strand and one newly synthesised strand.

They grew E. coli in a medium containing the heavy nitrogen isotope ¹⁵N so that all its DNA became 'heavy'.
