Both somatic hybridization and somaclonal variation are made possible by tissue culture, but they arise very differently from each other, and both differ fundamentally from classical, seed-based cross-hybridization.

Classical cross-hybridization requires the two parent plants to be close enough relatives that their pollen and egg cells can fuse and produce viable, fertile seed -- a sexual-compatibility barrier that rules out combining useful traits from more distantly related species altogether, however desirable those traits might be. Somatic hybridization deliberately gets around this barrier by working with body (somatic) cells instead of sex cells. The rigid cellulose cell wall is enzymatically digested away from cells of each parent, leaving naked protoplasts; protoplasts from the two parents -- which may belong to different species or even different genera -- are brought together and fused, typically using polyethylene glycol or a brief electric pulse, forming a single hybrid cell carrying the combined genetic material of both parents. This hybrid cell is cultured, regenerates a new cell wall, divides into a callus, and is eventually regenerated into a complete somatic hybrid plant -- a genuinely new plant type combining traits from two species ordinary breeding could never have crossed.
