Both Down and Klinefelter syndromes arise from non-disjunction (failure of chromosome separation) during meiosis, producing an extra chromosome; the kinetochore is the centromeric site of spindle-fibre attachment.

Reason for Down Syndrome: Caused by non-disjunction of chromosome 21 during gamete formation (meiosis), so a gamete carries two copies of chromosome 21 instead of one. Fertilisation with a normal gamete results in an individual with three copies of chromosome 21 (Trisomy 21) - total 47 chromosomes. Features include short stature, small round head, furrowed tongue, and partial mental retardation.

Reason for Klinefelter Syndrome: Caused by non-disjunction of the sex chromosomes, producing a gamete with an extra X chromosome. Fertilisation gives an individual with karyotype 47, XXY. Affected individuals are phenotypically male but show feminine body development (gynecomastia), sterility, and low intelligence.
