Micro-propagation exploits totipotency: a small explant grown in vitro on a suitable nutrient medium can be induced to regenerate thousands of genetically identical, disease-free plantlets in a short time and small space.

Micro-propagation is a plant tissue-culture technique in which a small piece of tissue (explant) is cultured on an artificial nutrient medium (containing sugars, minerals, vitamins and plant growth regulators such as auxins and cytokinins) under aseptic conditions, and induced — via a totipotent, undifferentiated callus — to regenerate into complete plantlets, which are then hardened and transferred to soil.

Advantages:

  1. Rapid, large-scale clonal multiplication — from a single explant, thousands of plants can be raised in a fraction of the time taken by conventional seed/vegetative propagation.
  2. Genetically identical (true-to-type) plants — since propagation is clonal/asexual, all the daughter plants retain the exact desirable traits of the selected parent, unlike seed propagation where segregation can alter traits.
  3. Disease-free plants — using meristem-tip culture (the shoot apical meristem is usually free of viruses), virus-free planting material can be produced even from an infected parent stock.
  4. Very little starting material needed — a tiny explant is sufficient, so even rare, endangered, or elite parent plants can be multiplied without destroying them. …
