Plant tissue culture exploits totipotency — the capacity of a single plant cell (even a fully differentiated somatic cell) to give rise to a complete plant, if provided the appropriate nutrient medium containing sugars, amino acids, vitamins, and growth regulators (auxins and cytokinins). A single cell (explant) is grown into an undifferentiated mass called a callus, which is then induced to differentiate into a complete plantlet — this is called somatic embryogenesis.
Applications:
- Micropropagation: Thousands of genetically identical plants (clones) can be produced in a short time and small space from a single parent plant — commercially used for ornamentals, fruit plants, and forest trees. This gives rapid multiplication with less time/labour than conventional propagation.
- Production of virus-free plants: In a plant infected by a virus, the meristem (apical/shoot tip) is usually virus-free even if the rest of the plant is infected. Culturing just this meristem tissue produces virus-free plants.
- Germplasm conservation: Rare, endangered, or economically valuable plant genetic material can be conserved for long periods at low temperature/growth as tissue-culture stocks (cryopreservation of cultured cells/embryos) without needing large field gene banks.
- Somaclonal variation: Tissue culture can generate genetic variants (somaclonal variants) useful in crop-improvement programmes for traits like disease resistance. …
