Tissue culture grows cells/tissues on artificial nutrient medium in sterile, in vitro conditions, exploiting the totipotency of plant cells; it has wide applications in agriculture, horticulture and research.

Tissue culture is defined as the aseptic (sterile) culture of cells, tissues, organs, or even whole organisms under controlled nutritional and environmental conditions, often in glass containers (hence also called in vitro culture). It relies on the property of totipotency — the inherent capacity of a living plant cell to give rise to a whole new organism when grown under suitable conditions, since a mature, differentiated cell can dedifferentiate and then redifferentiate to regenerate the entire plant.

Applications of tissue culture:

  1. Micropropagation: rapid clonal (vegetative) multiplication of large numbers of genetically identical, disease-free plantlets from a small piece of tissue — widely used for ornamental plants, fruit trees, and other commercially valuable species.
  2. Production of virus-free plants: culturing the shoot apical meristem (which is usually free of viruses even in an infected plant) to regenerate healthy, virus-free plants.
  3. Somatic hybridisation: fusing protoplasts (cell-wall-removed cells) of two different plant species/varieties in vitro to produce a hybrid plant that could not otherwise be obtained by normal sexual crossing (e.g. the tomato–potato hybrid 'pomato'). …
