Tissue culture grows isolated cells/tissue in sterile artificial medium, exploiting totipotency, and is applied to mass-clone plants, fuse species, and preserve germplasm.

Tissue culture is the in-vitro technique of growing cells, tissues, or organs of a plant (or animal) in a suitable artificial nutrient medium under sterile, controlled conditions. It relies on the property of totipotency — the inherent capacity of a living plant cell to regenerate a whole new organism, first demonstrated by Haberlandt. In plants, when this is used to produce thousands of genetically identical plantlets in a short time from a single cell/tissue explant, it is called micropropagation.

Applications:

  1. Micropropagation — rapid, large-scale multiplication of plants to produce thousands of genetically identical (clonal) progeny in a short duration and limited space; used commercially to raise disease-free plants of banana, sugarcane, apple, and many ornamental species.
  2. Somatic hybridization — protoplasts (cells with the wall removed) from two different plant varieties/species are fused (using PEG or electrofusion) to produce a hybrid protoplast, which can be cultured into a new hybrid plant, bypassing sexual incompatibility between distant species (e.g. the pomato, a somatic hybrid of potato and tomato). …
