The carbylamine (isocyanide) reaction converts a primary amine into a foul-smelling isocyanide when heated with chloroform and alcoholic KOH; because secondary and tertiary amines do not react this way, it serves as a specific qualitative test for primary amines.

The reaction. A primary amine (aliphatic or aromatic), when warmed with chloroform and alcoholic potassium hydroxide, is converted into an extremely unpleasant-smelling isocyanide (carbylamine):

R–NH2+CHCl3+3 KOH→  Δ  alcoholicR–NC  (isocyanide)+3 KCl+3 H2OR\text{–}NH_2 + CHCl_3 + 3\,KOH \xrightarrow[\;\Delta\;]{\text{alcoholic}} R\text{–}NC \;(\text{isocyanide}) + 3\,KCl + 3\,H_2O

(Mechanistically, alcoholic KOH generates dichlorocarbene, :CCl2:CCl_2, from chloroform, which is attacked by the amine's nitrogen lone pair; successive loss of HClHCl gives the isocyanide.)
