The carbylamine reaction is a characteristic, highly sensitive chemical test that identifies primary amines by the extremely unpleasant smell of the isocyanide (carbylamine) produced.

When a primary amine (aliphatic, R−NH2R-NH_2, or aromatic, Ar−NH2Ar-NH_2) is heated with chloroform (CHCl3CHCl_3) and ethanolic potassium hydroxide, an isocyanide (carbylamine) is formed, having a distinctly offensive odour:

R−NH2+CHCl3+3KOH→ΔR−NC+3KCl+3H2OR-NH_2 + CHCl_3 + 3KOH \xrightarrow{\Delta} R-NC + 3KCl + 3H_2O

Mechanistically, KOH first generates dichlorocarbene (:CCl2:CCl_2) from chloroform, which is attacked by the amine's nitrogen lone pair; two successive eliminations of HClHCl (mediated by more KOH) then give the isocyanide.
