The carbylamine (isocyanide) reaction is a diagnostic test for primary amines: heating with chloroform and alcoholic potassium hydroxide produces a foul-smelling alkyl/aryl isocyanide.

When a primary amine (aliphatic or aromatic) is warmed with chloroform in the presence of alcoholic potassium hydroxide, it is converted into an extremely foul-smelling isocyanide (carbylamine):

R−NH2+CHCl3+3KOH (alc.)→ΔR−N−≡C++3KCl+3H2OR-NH_2 + CHCl_3 + 3KOH\ (alc.) \xrightarrow{\Delta} R-\overset{-}{N}\equiv \overset{+}{C} + 3KCl + 3H_2O

(Mechanistically, alcoholic KOH first generates dichlorocarbene, :CCl2:CCl_2, from chloroform, which is attacked by the primary amine's nitrogen; successive loss of two HCl molecules under the strongly basic conditions gives the isocyanide.)

A specific example, with methylamine:

CH3NH2+CHCl3+3KOH→ΔCH3NC+3KCl+3H2OCH_3NH_2 + CHCl_3 + 3KOH \xrightarrow{\Delta} CH_3NC + 3KCl + 3H_2O
