Steps involved:
- Selection of ingredients: nutrients are chosen based on the nutritional requirements of the organism to be cultured - a carbon/energy source (peptone, beef extract, yeast extract, or sugars), nitrogen source, mineral salts, and, for solid media, a gelling agent (agar).
- Weighing and dissolving: the ingredients are weighed accurately on a balance and dissolved in the required volume of distilled water, heating gently with stirring to ensure complete dissolution.
- pH adjustment: the pH of the medium is checked with a pH meter or indicator paper and adjusted to the optimum range for the target organism (usually about 7.0-7.4 for most bacteria) using dilute HCl or NaOH.
- Adding the gelling agent: for solid media, agar (1.5-2%) is added and the mixture is boiled until it dissolves completely and the medium turns clear.
- Dispensing: the molten medium is distributed into test tubes, flasks, screw-cap bottles, or conical flasks in the required quantities, plugged with cotton or capped loosely to permit steam penetration.
- Sterilisation: the dispensed medium is sterilised, most commonly by autoclaving at 121°C and 15 lb/in² pressure for 15-20 minutes, which kills all vegetative cells, spores and other contaminants. …
