Photorespiration is a light-dependent, energy-wasting side reaction of RuBisCO involving three organelles, while dark respiration is the everyday, energy-yielding oxidation of respiratory substrates in the mitochondria — they differ in substrate, timing/organelles, and energy outcome.

Three clear differences between photorespiration and dark (cellular) respiration:

  • Photorespiration begins when the enzyme RuBisCO acts as an OXYGENASE on RuBP (ribulose-1,5-bisphosphate) instead of its normal substrate CO2, producing one molecule of phosphoglycolate (a 2-carbon compound) alongside 3-phosphoglycerate.

  • Dark respiration uses glucose (or other stored carbohydrates/respiratory substrates) as its starting substrate, breaking it down through glycolysis, the Krebs cycle, and oxidative phosphorylation.

  1. Occurrence (light dependence and organelles involved):
  • Photorespiration occurs ONLY IN THE LIGHT, since it depends on RuBisCO's oxygenase activity being favoured under high O2/low CO2 and high light/temperature conditions; its reactions span three organelles in sequence — chloroplast, peroxisome, and mitochondrion.

  • Dark respiration occurs CONTINUOUSLY, both day and night, since it does not depend on light; it mainly takes place in the cytoplasm (glycolysis) and mitochondria (Krebs cycle and electron transport chain).
