For blood cell counting, RBCs are diluted using Hayem's fluid, while WBCs are diluted using Turk's fluid — each formulated to make the target cell type countable under the microscope.

When counting blood cells using a hemocytometer, the blood sample must first be diluted with a suitable fluid to spread the cells out and, in the case of the WBC count, to destroy/lyse the far more numerous RBCs so they don't obscure the WBCs:
