A biochemical mutation is a mutation that disrupts a gene coding for an enzyme in a specific biosynthetic pathway, so the mutant organism (an auxotroph) can no longer synthesise an essential metabolite (amino acid, vitamin, etc.) on its own and must be supplied it externally to survive/grow, unlike the wild type (prototroph) which can synthesise it from simple starting materials.
