Both missense and nonsense mutations are types of point mutations (single base-pair substitutions) that alter one codon in the DNA/mRNA sequence, but they affect the resulting protein very differently.
A missense mutation occurs when the base change alters a codon so that it now codes for a different amino acid than the one originally specified. The resulting protein is still full length, but carries one wrong amino acid at that position. Depending on where the substitution occurs and how chemically similar the substituted amino acid is, the protein's function may be unaffected, reduced, or lost entirely. A well-known example is the mutation in the beta-globin gene that replaces glutamic acid with valine, producing abnormal haemoglobin and causing sickle-cell anaemia.
