Step 1. Recall the three named PCR steps from the chapter: denaturation, renaturation/primer annealing, and synthesis/primer extension. Step 2. Denaturation must come first — high heat (~95°C) is needed to split the double-stranded template DNA into single strands before anything else can happen. Step 3. Annealing comes second — the mixture is cooled so the primers can bind (hybridise) to their complementary sequences on the now-single-stranded template; this cannot happen before denaturation exposes the single strands.
