Step 1. All nucleic acids, including DNA, contain phosphorus as part of their phosphate backbone, but DNA (unlike protein) contains essentially no sulphur.

Step 2. Proteins, by contrast, contain sulphur in two of their amino acids, cysteine and methionine, but contain essentially no phosphorus at all.

Step 3. Because phosphorus is found only in the phage's DNA and sulphur is found only in the phage's protein coat, radioactively labelling each element separately let Hershey and Chase track DNA and protein as two completely independent, non-overlapping signals, with no risk of one isotope's radioactivity being mistaken for the other's. …
