In eukaryotic cells, the very long DNA molecule must be packaged to fit inside the nucleus. This is achieved with the help of positively charged, basic proteins called histones, which organise DNA (a negatively charged, acidic molecule) into a compact structure called chromatin. The nucleosome is the basic unit of this packaging.
Structure of a nucleosome:
- A histone octamer forms the core — this consists of two molecules each of four histone types: H2A, H2B, H3 and H4.
- Approximately 200 base pairs of DNA are wrapped around this histone octamer (about 1.65 to nearly 2 turns), forming the nucleosome core particle.
- Nucleosomes are connected to each other by a stretch of linker DNA.
- A fifth histone, H1, sits at the point where DNA enters and exits the nucleosome, helping to seal/organise the DNA around the core and stabilising higher-order packing.
