Pyruvate is made in the cytoplasm by glycolysis, and pyruvate dehydrogenase later converts it to acetyl CoA in the mitochondrial matrix.

The site where pyruvate is first synthesised is the cytoplasm of the cell, where glycolysis takes place. Glucose (derived from sucrose or storage carbohydrates) is broken down through a chain of ten enzyme-catalysed reactions into two molecules of pyruvic acid. This is true across all living organisms, since glycolysis is present universally.

Once formed, pyruvic acid can follow different fates depending on oxygen availability. Under aerobic conditions, it is transported from the cytoplasm into the matrix of the mitochondrion, where the enzyme pyruvate dehydrogenase catalyses its oxidative decarboxylation, working together with the coenzymes coenzyme A and NAD+.
