The 38-ATP net gain is a theoretical calculation valid only under four assumptions about an orderly, glucose-only pathway with no intermediates diverted and all glycolytic NADH oxidised in the mitochondria.

Calculating the net gain of ATP for each glucose oxidised can only be done as a theoretical exercise, and only if we make these assumptions:

  1. A sequential, orderly pathway is functioning, with one substrate forming the next, and with glycolysis, the TCA cycle and the ETS following one after another.
  2. The NADH synthesised in glycolysis is transferred into the mitochondria and undergoes oxidative phosphorylation there.
  3. None of the intermediates in the pathway is withdrawn to synthesise any other compound (i.e. the pathway is not tapped for biosynthesis).
  4. Only glucose is being respired — no other alternative substrate enters the pathway at any of the intermediary stages.
