Oxidative phosphorylation is the ETS-linked synthesis of ATP from ADP and inorganic phosphate, powered by the energy released as NADH and FADH2 are oxidised and electrons flow to oxygen.

Oxidative phosphorylation is the process by which ATP is synthesised from ADP and inorganic phosphate (Pi) using the energy liberated during the oxidation of NADH + H+ and FADH2 through the electron transport system (ETS).

How it works

  • The NADH and FADH2 formed in glycolysis and the Krebs' cycle are oxidised by the ETS carriers on the inner mitochondrial membrane.
  • As electrons pass from one carrier to the next and finally to oxygen (the final acceptor, forming water), energy is released.
  • This energy is used to pump protons across the inner membrane, setting up an electrochemical proton gradient.
  • The protons flow back through ATP synthase (complex V) down this gradient, and the enzyme couples this flow to the production of ATP from ADP and Pi (as explained by the chemiosmotic hypothesis).
