A cloning vector is a self-replicating carrier DNA (plasmid or phage) that ferries foreign DNA into a host and multiplies it; it needs an ori, a selectable marker and unique restriction sites.

What it is. In genetic engineering the gene of interest cannot enter and multiply in a host cell on its own. A cloning vector is a DNA molecule that carries the foreign DNA into the host cell and replicates within it, so that many identical copies (clones) of the inserted DNA are produced. Common vectors are plasmids (e.g. pBR322) and bacteriophages.

Essential features of a good cloning vector.

  1. Origin of replication (ori): the sequence from which replication starts; it allows the vector (and the linked foreign DNA) to multiply inside the host.
