Ori is the sequence that lets a vector self-replicate inside a host cell, producing multiple copies of the inserted foreign DNA.

For recombinant DNA technology to work, a foreign DNA fragment ('gene of interest') must be inserted into a vector (like a plasmid) that can replicate inside a host cell such as E. coli.
