(a) PCR amplifies a specific gene/DNA segment of interest without needing to clone it in a living cell. Each amplification cycle has three steps: first, denaturation, where the reaction mixture is heated to around 94-96C, separating the double-stranded template DNA into two single strands; second, annealing (primer binding), where the temperature is lowered (typically 50-65C) so that two short, sequence-specific single-stranded DNA primers can bind to their complementary sequences flanking the target region on each separated strand; and third, extension, where the temperature is raised again (around 72C) and a heat-stable DNA polymerase (commonly Taq polymerase, isolated from the thermophilic bacterium Thermus aquaticus, chosen because it survives the repeated high-temperature denaturation steps) extends each primer, synthesising a new complementary strand. Because each cycle roughly doubles the number of copies of the target sequence, repeating this three-step cycle 20-30 times produces millions of copies of the desired DNA segment from even a minute starting sample, which is the basis of PCR's use in cloning, diagnostics and forensics. (b) Ascariasis, caused by the intestinal roundworm Ascaris lumbricoides, is transmitted when a person ingests food or water contaminated with infective eggs; the resulting infection causes abdominal pain, digestive disturbance, malnutrition (as the worms consume nutrients), and in heavy infections, intestinal blockage.
