Because each single turn of the Calvin cycle fixes only one molecule of CO2, and glucose is built from six carbon atoms, exactly six turns of the cycle — fixing six CO2 molecules — are needed to assemble one molecule of glucose.

The Calvin cycle operates in three stages every time it turns: carboxylation, in which one molecule of CO2 is fixed onto RuBP by RuBisCO to yield two molecules of PGA; reduction, in which ATP and NADPH are used to convert this fixed carbon toward sugar; and regeneration, in which the CO2 acceptor RuBP is reconstituted using further ATP so the cycle can begin again. Each of these turns processes just one molecule of CO2, contributing just one carbon atom toward eventual sugar synthesis.

Glucose, the sugar that photosynthesis ultimately produces, is a 6-carbon molecule. Since a single turn of the cycle supplies only one carbon, it takes six separate turns — and therefore the fixation of six separate CO2 molecules — before enough carbon has been accumulated to be assembled into one molecule of glucose. …
