The carbylamine (isocyanide) reaction is a test for primary amines, converting them to foul-smelling isocyanides.

When a primary aliphatic amine is heated with chloroform (CHCl3) and alcoholic potassium hydroxide (KOH), it is converted into an isocyanide (carbylamine) with a characteristic offensive smell. This reaction is used as a diagnostic test for primary amines.

Mechanism outline: alcoholic KOH first generates dichlorocarbene (:CCl2:CCl_2) from chloroform, which reacts with the free -NH2 group of the primary amine; subsequent loss of two more molecules of HCl (as KCl + H2O) gives the isocyanide.

Example – methylamine (a primary aliphatic amine):

CH3NH2+CHCl3+3KOH→ΔCH3NC+3KCl+3H2OCH_3NH_2 + CHCl_3 + 3KOH \xrightarrow{\Delta} CH_3NC + 3KCl + 3H_2O
