Every enzyme carries a specific pocket, the active site, into which its particular substrate can settle. The catalytic cycle unfolds in a clear sequence of steps once that first binding event has taken place:
- Binding - the substrate first diffuses towards the active site and docks into it, fitting into the specially shaped crevice or pocket, forming the transient enzyme-substrate (ES) complex.
- Induced fit and the transition state - the arrival of the substrate causes the enzyme to adjust its own shape, wrapping more snugly around the substrate. This close, strained embrace corresponds to the transition state, an unstable, high-energy arrangement in which the substrate's structure is being distorted towards that of the product.
- Bond breaking and product formation - held tightly and very close to the enzyme's reactive groups, the substrate has its chemical bonds broken, or new bonds made, and this converts the arrangement into an enzyme-product (EP) complex. …
