Carrying an explant through to a regenerated plantlet is not a single operation but a sequence of linked techniques, each covered as its own subsection below: sterilization of everything the explant will contact, preparation of the nutrient medium, setting the physical culture conditions, inducing callus formation, allowing embryogenesis (or, further on, organogenesis), culturing and fusing protoplasts where relevant, and finally hardening the resulting plantlet for life outside the culture vessel.
