Q.Hershey and Chase experiment with bacteriophage showed that
Step 1. T2 phage grown with radioactive phosphorus-32 had labelled DNA (DNA is rich in phosphorus); phage grown with radioactive sulphur-35 had labelled protein (protein is rich in sulphur, DNA has none).
Step 2. After each labelled batch infected unlabelled E. coli, blending sheared off any phage material still stuck to the outside of the bacteria, and centrifugation separated the bacterial pellet from the phage debris in the supernatant.
Step 3. Radioactivity from 32P (DNA) was found inside the bacterial pellet; radioactivity from 35S (protein) stayed in the supernatant and never entered the cells. Even the next phage generation carried only 32P.
Because only the DNA physically entered the bacterium and directed synthesis of new phage, the experiment demonstrated that (b) DNA is the genetic material.
