Step 1. Substrate concentration: raising substrate concentration raises reaction velocity up to a maximum, Vmax, at which all active sites are saturated; the substrate concentration giving half-maximal velocity is the Michaelis-Menten constant, Km — a low Km indicating high enzyme-substrate affinity, a high Km indicating low affinity.

Step 2. Enzyme concentration: with substrate in excess, reaction rate rises as enzyme concentration increases, since more active sites become available.

Step 3. Temperature: activity peaks at an optimum temperature (around 37°C in homeotherms); activity rises with temperature only up to this point, and enzymes denature rapidly above about 40°C.
