Benzene and PAHs (e.g. benzo[a]pyrene, produced whenever an organic fuel burns incompletely -- tobacco smoke, vehicle exhaust, charred food) are, in their unmodified form, too chemically stable and non-polar to react directly with biological molecules such as DNA. The danger instead arises because the body's own liver enzymes -- cytochrome P450 enzymes, whose normal job is to oxidise fat-soluble foreign compounds to make them more water-soluble and excretable -- oxidise the aromatic ring and convert it into a reactive epoxide intermediate. Rather than being harmlessly excreted, this epoxide IS chemically reactive and electrophilic, and it can bond covalently to nucleophilic sites on DNA (most classically, the amino group of a guanine base), forming a DNA adduct. If this adduct is not correctly repaired by the cell before the DNA is next replicated, it can cause a permanent mutation; an accumulation of such mutations, particularly in genes controlling cell growth and division, is a well-established route to cancer -- benzo[a]pyrene is one of the best-studied chemical carcinogens acting by exactly this mechanism, strongly associated with lung and skin cancers.
