Endonucleases (restriction enzymes) cut DNA within the strand at a specific short palindromic sequence called the restriction site.

Endonuclease: A type of nuclease enzyme that cleaves the phosphodiester bond WITHIN a DNA/RNA strand (at internal positions), as opposed to an exonuclease, which removes nucleotides one at a time from the ends of the strand. Restriction endonucleases are a special class that recognise and cut DNA only at a specific nucleotide sequence.
