Genetic engineering follows a set sequence - isolate the gene, splice it into a vector, transform a host cell, then select and express it; cDNA is the DNA copy of an mRNA made using reverse transcriptase.

Steps in Genetic Engineering:

  1. Isolation of genetic material (DNA/gene of interest): the desired gene is identified and isolated, often from genomic DNA or synthesised from mRNA as cDNA.
  2. Cutting (restriction digestion) and Ligation: the gene of interest and a vector (plasmid/phage) are cut using the same restriction enzyme, producing complementary sticky ends; DNA ligase joins them to form recombinant DNA (rDNA).
  3. Transfer of rDNA into the host cell (transformation): the recombinant DNA is introduced into a suitable host cell (bacterium, plant, or animal cell) via methods like heat shock, electroporation, or biolistics (gene gun).
  4. Selection of transformed host cells and multiplication/expression: using selectable markers (e.g., antibiotic resistance genes), transformed cells are identified, and the gene product (e.g., insulin) is expressed and harvested/downstream processed. …
