Step 1. The clearest structural difference is the presence of ribosomes: rough ER has ribosomes attached to its outer surface, giving it a studded appearance, while smooth ER has no ribosomes on its surface.

Step 2. This structural difference matches a functional one. Rough ER is chiefly involved in protein synthesis -- for example, in pancreatic cells secreting insulin -- with the resulting secretory proteins wrapped in membrane and budded off as transport vesicles from a transitional ER region; it also helps build new membrane for the cell.

Step 3. Smooth ER, lacking ribosomes, instead carries out a range of other jobs depending on the cell type: synthesising lipids/steroids (e.g. in the adrenal cortex, testes and ovaries), detoxifying drugs and poisons (in liver cells), and storing calcium ions (in muscle cells). …
