PCR amplifies DNA by cycling through denaturation, annealing and extension.

Polymerase Chain Reaction (PCR) is an in-vitro technique to amplify a specific DNA segment millions of times, using a thermal cycler. Each cycle has three steps: (1) Denaturation — the reaction mixture is heated to about 94-96 degC, which breaks the hydrogen bonds and separates the double-stranded DNA template into two single strands. (2) Annealing — the temperature is lowered to about 50-65 degC, allowing two short, synthetic, single-stranded primers (complementary to sequences flanking the target region on each strand) to bind (anneal) to their matching sites.
