Q.In a PCR machine the temperature can be raised up to 100 degrees C but after that it is not able to lower the temperature below 70 degrees C automatically. Which step of PCR will be hampered first in this faulty machine? Explain why.

Step 1. A PCR cycle has three temperature-dependent steps: denaturation (90-98°C), annealing (40-60°C), and primer extension (70-75°C).

Step 2. The faulty machine can reach up to 100°C but cannot go below 70°C -- so denaturation (needing up to 98°C) still works fine, since 100°C comfortably covers that range.

Step 3. Primer extension, which needs 70-75°C, would also still work, since the machine's lower limit of 70°C is right at the bottom of this range.

Step 4. Annealing, however, needs the mixture to cool to 40-60°C so the primers can pair with their complementary sequences on the single-stranded DNA -- at 70°C and above, the primers cannot bind stably to the template. …
