The chapter states a vector must contain an origin of replication (ori), 'a sequence from where replication starts', so that any DNA linked to it is replicated along with the vector inside the host cell.
Step 2. Check for a selectable marker. A vector should carry a suitable marker, such as antibiotic resistance, that permits detection of transformed host cells - identifying which cells actually took up the vector.
