Southern blotting separates and detects DNA; Northern blotting separates and detects RNA; Western blotting separates and detects proteins - this one difference is the root of almost everything else that differs between them.
Step 2. Compare the naming. Southern blotting is genuinely named after its inventor, E.M. Southern; 'Northern' and 'Western' are simply plays on that name, not references to any actual place or person.
Step 3. Compare denaturation needs. Southern and Western blotting both require a denaturation step (unwinding double-stranded DNA, or unfolding protein) before transfer; Northern blotting does not need this step.
Step 4. Compare the membrane used. Southern and Western blotting both typically use nitrocellulose or nylon membrane; Northern blotting instead uses amino benzyloxymethyl paper, specifically because RNA does not bind well to plain nitrocellulose. …
