The problem with correcting only mature lymphocytes, as explained in the previous question, is that mature lymphocytes are short-lived and are not capable of self-renewal, so any corrective benefit provided by a batch of corrected mature lymphocytes is inherently temporary and must be periodically renewed by repeating the entire treatment procedure.

Bone-marrow lymphocyte stem cells are fundamentally different in this respect: they are capable of long-term self-renewal (dividing to produce more stem cells, maintaining their own population over an entire lifetime) while also continuously giving rise to new, mature lymphocytes through ongoing differentiation, replenishing the body's supply of lymphocytes throughout life without needing to be reintroduced from outside. …
