Step 1. Independent replication: a good vector must be able to replicate on its own inside the host cell, through its own origin-of-replication (ori) gene, so that as it copies itself, many copies of the inserted foreign DNA are produced too.

Step 2. Easy host entry: the vector must be able to get into host cells readily, so that transformation is efficient.

Step 3. Selectable marker genes: a good vector carries marker genes, typically for antibiotic resistance, so that cells which have taken up the vector (and, further, the recombinant vector) can be distinguished from those that have not.

Step 4. Unique cleavage site: it must contain a unique restriction-enzyme cleavage site located inside one of its marker genes, so that inserting foreign DNA there disrupts that one marker (letting recombinant cells be told apart from non-recombinant ones, as with PstI and the ampicillin-resistance gene of pBR322). …
