Step 1 -- Enzymes. Several enzymes are used in rDNA technology, including lysozymes, nucleases (exonucleases, endonucleases, restriction endonucleases), DNA ligases, DNA polymerases, alkaline phosphatases and reverse transcriptases. Restriction endonucleases specifically cut DNA at defined recognition sequences to release the desired gene and open up the vector, while DNA ligase then joins the foreign DNA into the cut vector to form recombinant DNA.

Step 2 -- Cloning vectors (vehicle DNA). These are DNA molecules -- most commonly plasmids (pBR322, pUC, the Ti plasmid) or bacteriophages (lambda, M13), but also cosmids, phagemids, BACs, YACs, transposons, baculoviruses or MACs -- that carry the foreign DNA segment and replicate inside a host cell. A good vector replicates independently, enters host cells easily, and carries marker genes plus control elements needed for later selection and expression. …
