Step 1. In standard rDNA technology, the same restriction enzyme is used to cut both the donor DNA (to release the desired gene) and the vector DNA (to open it up for insertion), so that both pieces end up with matching, complementary sticky (or blunt) ends.

Step 2. Different restriction enzymes generally recognise different sequences and can produce different types of cut end -- for example, one enzyme might leave a sticky end with a particular single-stranded overhang, while another leaves a blunt end or a differently-shaped overhang.
