A cloning vector (e.g. a plasmid such as pBR322) is identified/characterised by three essential features:
- Origin of replication (ori): a specific DNA sequence from which replication starts, allowing the vector (and any DNA inserted into it) to replicate autonomously and be maintained inside the host cell, independent of the host chromosome. The ori also controls the copy number of the vector per cell.
- Selectable marker gene: a gene, commonly conferring resistance to an antibiotic (e.g. ampicillin or tetracycline resistance in pBR322), which allows host cells that have taken up the vector (transformants) to be distinguished and selected from those that have not (non-transformants), typically by growing cells on a medium containing that antibiotic. …
