DNA supercoiling refers to the over- or under-winding of the double helix relative to its normal relaxed state. Positive supercoiling occurs when DNA is overwound (twisted further in the same sense as the helix), which happens naturally ahead of an advancing replication fork or transcription bubble as the two strands are unwound, creating torsional strain in the DNA ahead of the moving enzyme complex.
